Alpha-2 adrenergic receptors are G protein-coupled receptors for catecholamines that activate G(i/o) protein pathway, thereby promoting adenylyl cyclase inhibition, ERK1/2 stimulation, and voltage-gated calcium channels suppression (PubMed:11056163, PubMed:2164221, PubMed:2172775, PubMed:23105096, PubMed:26811329). Control a variety of physiological processes, such as regulation of blood pressure, lipolysis and insulin release (PubMed:2164221). The rank order of potency for agonists of ADRA2B is clonidine > norepinephrine > epinephrine = oxymetazoline > dopamine > p-tyramine = phenylephrine > serotonin > p-synephrine / p-octopamine.
For antagonists, the rank order is yohimbine > chlorpromazine > phentolamine > mianserine > spiperone > prazosin > alprenolol > propanolol > pindolol (PubMed:2164221, PubMed:2172775)
Interacts with RAB26; RAB26 mediates cell surface transport from the Golgi (PubMed:23105096). Interacts with PPP1R9B (PubMed:24114805). Interacts with GGA1, GGA2 and GGA3; GGA1, GGA2 and GGA3 mediates transport from the Golgi to the cell membrane (PubMed:26811329, PubMed:27901063).
Interacts with RAB43 (GTP-bound); RAB43 mediates ADRA2B ER-to-Golgi cell transport (PubMed:29069590)
A form of familial myoclonic epilepsy, a neurologic disorder characterized by cortical hand tremors, myoclonic jerks and occasional generalized or focal seizures with a non-progressive or very slowly progressive disease course. Usually, myoclonic tremor is the presenting symptom, characterized by tremulous finger movements and myoclonic jerks of the limbs increased by action and posture. In a minority of patients, seizures are the presenting symptom.
Some patients exhibit mild cognitive impairment. FAME2 inheritance is autosomal dominant.

Genes with an experimentally identified or computationally predicted synthetic-lethal relationship to ADRA2B, aggregated across our SSL data sources. Click any partner node to view that gene’s page.
Nodes and edges are coloured by the SSL data source. Partners appearing in more than one source are shown in grey.
